Enterokinase specifically hydrolyzes peptide bond at the carboxyl side of lysine residue preceded by four aspartic acids: Asp-Asp-Asp-Asp-Lys (DDDDK). So, Enterokinase can remove N-terminal fusion protein or tags to get aim protein with native amino acids sequence.
