Description

 

Specificity and Kinetics Trypsin will cleave peptides on the C-terminal side of lysine and arginine amino acid residues.

The rate of hydrolysis is slower if an acidic residue is on either side of the cleavage site and no cleavage occurs if a proline residue is on the carboxyl side of the cleavage site.Trypsin will hydrolyze ester and amide linkages of several synthetic substrates.The pH optimum of trypsin is 7 - 10.Assay Method The activity of most Sigma preparations is determined by a continuous rate spectrophotometric assay and expressed in BAEE units.

 

Unit Definition: One BAEE unit will produce a ΔA253 of 0.001 per min at pH 7.6 at 25 °C using

BAEE as substrate. Reaction volume = 3.2 mL (1 cm light path).

Temp = 25 °C, pH = 7.6, A253nm, Light path = 1 cm
In a 3.2 ml reaction mix, the final concentrations are 63 mM sodium phosphate, 0.23 mM Nα-benzoyl-L-arginine ethyl ester, 0.06 mM hydrochloric acid, and 100 units trypsin.
